But many bind to other receptors too, producing side effects that only show up after tests in animals or people. [...] Using published databases of interactions between small molecules, or "ligands", and potential targets, they used software to search for similarities in chemical structure between ligands and 3665 drugs either in use or under development. This yielded a list of nearly 4000 previously unknown potential targets. Some of these new targets might explain known side effects. The software predicted that the anti-nausea drug domperidone should also bind to a receptor that contributes to cardiac arrhythmia – one of the drug's side effects. Others point to new uses: the antihistamine mebhydrolin should block a receptor involved in Alzheimer's disease. [...]

UNAIDS, the Joint United Nations Programme on HIV/AIDS, is an innovative joint venture of the United Nations family, bringing together the efforts and resources of ten UN system organizations in the AIDS response to help the world prevent new HIV infections, care for people living with HIV, and mitigate the impact of the epidemic.
With its headquarters in Geneva, Switzerland, the UNAIDS Secretariat works on the ground in more than 80 countries worldwide. Coherent action on AIDS by the UN system is coordinated in countries through the UN theme groups, and the joint programmes on AIDS.

Scientists have found a way to disarm a protein thought to play a key role in leukaemia and other cancers. [...] Previous attempts to neutralise the protein had failed, leading experts to conclude it was effectively "undruggable". [...] They employed a state-of-the-art technique using chemical braces to mould protein snippets called peptides into specific three dimensional shapes. These "stapled" peptides are readily absorbed by cells, and are so tiny they can be deployed to alter gene regulation at specific sites. [...] When tested in mice it was found to limit the growth of cancer cells. [...] Researcher Professor Greg Verdine said: "Stapled peptides promise to significantly expand the range of what's considered 'druggable'. "With our discovery, we've declared open season on transcription factors and other intractable drug targets." [...]

Vaccines on Horizon for AIDS, Alzheimer's, Herpes

'Golden era' could bring vaccines against AIDS, Alzheimer's and addictions within 5 years

Malaria. Tuberculosis. Alzheimer's disease. AIDS. Pandemic flu. Genital herpes. Urinary tract infections. Grass allergies. Traveler's diarrhea. You name it, the pharmaceutical industry is working on a vaccine to prevent it.
Many could be on the market in five years or less.
Contrast that with five years ago, when so many companies had abandoned the vaccine business that half the U.S. supply of flu shots was lost because of contamination at one of the two manufacturers left....

Some new research suggesting which protein to aim any HIV vaccine or attachment blocking research at:

New findings suggest strategy to help generate HIV-neutralizing antibodies

Suggests that it will need to aim specifically at trimers of this protein, and in ways that don't break up the trimer, to have much effect.
